Abstract

The low production of garlic (Allium sativum L.) in Indonesia has encouraged the development of tissue culture as an alternative propagation method. Successful callus induction is influenced by the source of the explant and the composition of plant growth regulators (PGRs). This study aimed to evaluate the effects of shoot parts and plant growth regulator compositions on in vitro callus induction of garlic. The experiment was arranged in a factorial Completely Randomized Design (CRD) with two factors: shoot parts and plant growth regulator compositions. The results showed a significant interaction between shoot parts and plant growth regulator compositions on the time required for callus initiation, the percentage of explants forming callus, and the percentage of embryogenic callus. The lower shoot explants cultured on medium supplemented with 1 mg L⁻¹ 2,4-D and 0.5 mg L⁻¹ BAP exhibited the best response for callus induction. The resulting calli were predominantly friable and yellowish in color, indicating their potential embryogenic characteristics based on morphological observations.
