Abstract

Indonesia has a wide variety of superior local banana cultivars, including kepok banana, raja banana, tanduk banana, barangan banana, and many others. This study aimed to determine the effect of different sterilization methods on the success of in vitro initiation of local Bogor kepok banana explants. The research was conducted at the Laboratory of the Tropical Horticulture Study Center (PKHT) in Bogor from April to June 2026. This study used a Completely Randomized Design (CRD) with one factor, namely the sterilization method (S). Four sterilization methods were applied: S1 (Streptomycin sulfate + benomyl (18 hours) and NaOCl + Tween 80 (30 minutes)), S2 (Streptomycin sulfate + benomyl (18 hours), NaOCl (30 minutes), and Tween 80 (30 minutes)), S3 (Streptomycin sulfate + benomyl (5 hours) and NaOCl + Tween 80 (30 minutes)), and S4 (Streptomycin sulfate + benomyl (5 hours), NaOCl (30 minutes), and Tween 80 (30 minutes)). The results showed that the sterilization method significantly affected the time to first contamination, the percentage of dead explants, and the percentage of surviving explants; however, it did not significantly affect the percentage of contamination, browning, and budding explants. Method S2 (Streptomycin sulfate + benomyl (18 hours), NaOCl (30 minutes), and Tween 80 (30 minutes)) proved to be the most effective sterilization method, producing sterile explants and achieving a budding explant percentage of 8.33%.
