Abstract

This study aims to examine the kinetics of cellulose hydrolysis into glucose in pineapple peel waste (Ananas comosus) using cellulase enzymes in the context of household organic waste processing. Pineapple peel waste has a fairly high cellulose content (61.88%) after going through a delignification process with 5% NaOH solution at 80°C for 90 minutes, which aims to increase the availability of cellulose substrates by removing lignin that inhibits enzyme access. Enzymatic hydrolysis was carried out at various substrate concentrations of 1% to 5% (% w/v) and hydrolysis times of 60 to 180 minutes at optimum conditions of pH 5 and 50°C using commercial cellulase enzymes. Measurement of hydrolysis results in the form of glucose concentration was carried out using a refractometer and analyzed using the Michaelis-Menten kinetic model. The results showed that glucose concentration increased significantly with increasing time and substrate concentration until it reached enzyme saturation. The highest maximum reaction rate (k) was achieved at a substrate concentration of 5% with a reaction rate equation of -r_A= r_R=0,3818 (C_E0 C_A)/(2,357+ C_A ), which indicates optimal efficiency of cellulose conversion. This study provides an important contribution to the utilization of household organic waste from pineapple peels as a source of glucose through an environmentally friendly enzymatic bioconversion process with potential for biofertilizer and bioenergy applications.
