Abstract
Brown rice has potential as a functional food due to its content of γ-aminobutyric acid (GABA); however, its natural GABA levels are relatively low, and the germination process is susceptible to microbial contamination. This study aimed to evaluate a partial circulation germination system equipped with a microfiltration membrane and ultraviolet (UV) irradiation to enhance GABA accumulation, preserve sprout quality, and control microbial communities. A two-factor completely randomized design was used, with UV treatment (UV and non-UV) and germination duration (24, 48, and 72 hours) as factors. Parameters assessed included germination percentage, sprout length, germination medium pH, GABA content, total microbial and lactic acid bacteria (LAB) counts using standard plate count methods, and microbial diversity using next-generation sequencing (NGS). Results showed that germination duration significantly increased GABA accumulation up to 48 hours. UV treatment did not significantly affect GABA levels but was effective in reducing total microbial counts during the early germination phase; however, its inhibitory effect declined with extended incubation. Metagenomic analysis of the 48-hour non-UV treatment revealed dominance of the genera Phytobacter and Enterobacter, which support seedling growth. The opportunistic pathogen Cronobacter was also identified, indicating its potential use as a sanitation indicator. This study offers an initial insight into the development of germination technologies and highlights the potential of metagenomic analysis for monitoring microbial dynamics and sanitation during the germination process.
