Abstract
Spodoptera exigua is a major pest of shallots (Allium ascalinicum) in East Java, capable of reducing yields by up to 45%. Farmer’s current control practice still rely on chemical insecticides, necessitating environmentally friendly control. This study aimed to determine the most effective concentrations of Bacillus isolate Bcz 16 and Bcz 20 in controlling Spodoptera exigua, the bacterial mechanism of action when infecting larva, the active compound produces and histopatological change in the midgut of infected larvae. The research was conducted from Januari to June 2026 at the laboratory of UPT Protection TPH East java and Laboratory of plant health Faculty of Agriculture UPN “Veteran” East Java, with HPLC dan FTIR analysis at Gadjah Mada University and histopatology test at Brawijaya University Malang. The research was arranged using a Comletely Randomized Design Factorial with factors of bacillus isolate type (Bcz 16 dan Bcz 20) and bacterial suspension concentration factors (30%, 40%, and 50%) which were repeated 5 time. The results of the study showed that Bcz 16 dan Bcz 20 isolated had a significant effect on larva mortality, pupa formation and imago formation, with a concentration of 40% as the most effective and efficient concentration. The highest mortality was produced by isolate Bcz 20 at 40.67% and Bcz 16 at 34%. HPLC analysis showed an increase in amino acid levels in larva infected with Bcz 16 and Bcz 20 by 150% and 165%, respectively, compared to helthy larvae, indicating impaired protein metabolism due to proteolytic activity and bacterial toxins. FTIR testing supported this finding by confirming the presence of extracellular compounds, endotoxins, and secondary metabolites in Bacillus isolate. Histopatologycal observations of the midgut of infected larvae showed cytoplasmic vacualation, epithelial cell swelling, and cell lysis, with severe damage in larvae infected with Bcz 20. Based on these result. Bacillus Bcz 16 and Bcz 20 have the potential to be developed as environmentally friendly bioinsecticides to control Spodoptera exigua in shallot. Keywords: Bacillus sp., Bioinsectiside, Histopatology, active compounds, Spodoptera exigua.
