Abstract
Fusarium wilt, known locally as moler disease, is a major constraint in shallot (Allium cepa L.) cultivation in the production centers of Probolinggo and Nganjuk, East Java. This study aimed to determine the morphology and type of Fusarium sp. infecting shallot plants in both locations through field observation, disease intensity calculation, isolation on potato dextrose agar (PDA), macroscopicmicroscopic morphological identification, and DNA amplification using polymerase chain reaction (PCR). Characteristic symptoms included leaf yellowing from the tip, twisting, and basal rot with brown to blackish discoloration. Mean disease intensity in Probolinggo reached 10.88%, considerably higher than in Nganjuk at only 0.49%. Three isolates (P1, NG2, NG3) displayed white, cottony colonies on PDA, with hyaline, branched, septate hyphae and abundant round to oval microconidia, whereas sickle-shaped macroconidia and championship were not observed. Agarose gel electrophoresis showed a single, consistent DNA band across all samples without double bands or smearing, indicating specific amplification suitable for sequencing. The morphological characters support genuslevel identification of Fusarium, while confirmation of the Fusarium oxysporum species still requires DNA sequencing data and comparison against the GenBank database.
