Abstract
This study optimizes glucose extraction from Saccharomyces cerevisiae regeneration waste via acid hydrolysis using H₂SO₄ and Response Surface Methodology (RSM). Key variables were waste volume (300–1500 mL) and water volume (200–1000 mL), with fixed conditions at 30°C, 180 min, and 200 rpm. Spectrophotometric analysis showed a peak glucose yield of 29.7% at 900 mL waste and 600 mL water. Minitab 19 optimization predicted 22.83% glucose at 1378.79 mL waste and 609.35 mL water (desirability: 0.929, R²: 95.57%). The results demonstrate the potential of using this waste for glucose production, supporting sustainable applications like bioethanol synthesis.
