Abstract
ugarcane bagasse has great potential as a raw material for bioethanol, but its conversion through enzymatic hydrolysis proceeds slowly and is costly. To overcome this, this study applied ultrasonic waves to accelerate the action of the cellulase enzyme, with the aim of investigating the effect of hydrolysis time and enzyme volume on the percentage of glucose produced. The method began with a delignification process using a 6% NaOH solution at 100°C for 1 hour. The lignin-free samples were then hydrolyzed at 40°C with the assistance of 40 kHz ultrasonication, using variations in enzyme volume (5-25 mL) and time duration (30-150 minutes). The sugar and glucose content data were subsequently optimized using Response Surface Methodology (RSM). The results showed that the delignification process successfully reduced the lignin content to 9.73% and increased the cellulose up to 57.33%. In the hydrolysis process, the utilization of ultrasonication consistently increased the total sugar by 20-30% compared to the conventional method. Experimentally, the highest efficiency was achieved using 25 mL of enzyme for 120 minutes, yielding 13.73% glucose and 15.4% total sugar (°Brix). Furthermore, the RSM modeling predicted the ideal optimum point to be at the use of 24.997 mL of enzyme for 149.828 minutes, which is projected to produce 13.734% glucose with a perfect desirability value (1.000).
