Abstract
This study examines the optimization of glucose levels from the liquid waste of Saccharomyces cerevisiae regeneration through hydrolysis using sulfuric acid (H₂SO₄) as a catalyst, employing Response Surface Methodology (RSM). The independent variables include waste volume (300–1500 mL) and water volume (200–1000 mL), with fixed conditions of temperature at 30°C, time at 180 minutes, and stirring at 200 rpm. The highest spectrophotometric measurement result reached 29.7% at a waste volume of 900 mL and water volume of 600 mL. Optimization using Minitab 19, based on 13 experimental data points, yielded an optimal glucose content of 22.83% at a waste volume of 1378.79 mL and water volume of 609.35 mL, with a desirability value of 0.929 and an R² of 95.57%. This research demonstrates that the hydrolysis process of liquid waste from Saccharomyces cerevisiae regeneration can be utilized to produce glucose as a value-added raw material, such as for bioethanol production.
