Abstract

The continuous decline in tomato crop production is caused by pest and plant disease attacks, one of which is the nematode Meloidogyne spp. Control is still carried out using synthetic nematicides that can have negative impacts on health, broad-spectrum effects, microorganism diversity, and resistance to Meloidogyne spp. nematode attacks. Therefore, there is a need for alternative control using biocontrol agents that are resistant to the presence of nematicide active ingredients (carbofuran and dazomet) and have the ability to inhibit Meloidogyne spp. nematode attacks. This study aims to test the resistance of Streptomyces spp. isolats to synthetic nematicide active ingredients, carbofuran and dazomet, and to evaluate their effect on Meloidogyne spp. nematode mortality. The method used was in vitro with the Poisoned Food technique and testing nematicidal activity against juvenile nematodes. The results showed that the TMP isolat had better resistance than SP against carbofuran, with the second-highest colony count of 11.47 × 10⁶ cfu/ml. Meanwhile, dazomet significantly inhibited the growth of Streptomyces spp. colonies. Mortality testing showed that the TMP isolat was able to achieve a mortality rate of Meloidogyne spp. of 59.01%. The best chitinolytic activity in Streptomyces spp. TMP isolat was 7.33 mm. Secondary metabolite analysis of Streptomyces spp. by headspace GC-MS of the TMP isolat produced volatile compounds with nematicidal, antibacterial, and antimicrobial activity. This proves the potential of Streptomyces spp. as a agent hayati in nematode management while being resistant to nematicide active ingredient residues, making it an environmentally friendly alternative in agricultural practices.
