Abstract
ABSTRAK Bawang putih (Allium sativum L.) termasuk salah satu tanaman hortikultura dan digunakan sebagai bumbu masak dan obat. Penelitian ini bertujuan untuk mendapatkan perbanyakan bawang putih melalui teknik kultur jaringan dengan penggunaan berbagai bagian eksplan tunas dan ukuran eksplan tunas bawang putih varietas Lumbu Putih. Penelitian dilaksanakan di Laboratorium Bioteknologi Fakultas Pertanian UPN “Veteran” Jawa Timur pada bulan Februari-Mei 2023. Penelitian merupakan percobaan faktorial dua faktor yang dirancang dalam Rancangan Acak Lengkap. Faktor pertama adalah bagian eksplan tunas bawang putih terdiri dari tunas bagian bawah, tengah dan atas. Faktor kedua yaitu ukuran eksplan tunas bawang putih terdiri dari ukuran 3 mm, 6 mm dan 9 mm. Hasil penelitian menunjukkan bahwa tidak terdapat interaksi antara bagian eksplan tunas dengan ukuran eksplan tunas bawang putih terhadap pertumbuhan planlet bawang putih. Tunas bagian bawah dengan ukuran 3 mm, 6 mm dan 9 mm mampu menginduksi planlet lengkap, tunas bagian tengah ukuran 3 mm, 6 mm dan 9 mm menghasilkan planlet lengkap, sedangkan eksplan bagian atas dengan ukuran 3 mm, 6 mm dan 9 mm belum mampu menunjukkan pertumbuhan planlet lengkap. Kata kunci: Pertumbuhan planlet, bagian eksplan tunas, ukuran eksplan tunas ABSTRACT Garlic (Allium sativum L.) is one of the horticultural crops and used as a cooking spice and medicine. This study aims to obtain garlic propagation through tissue culture techniques using various parts of the explant shoots and the size of the explants of the Lumbu Putih variety. The research was conducted at the Biotechnology Laboratory of the Faculty of Agriculture UPN "Veteran" East Java in February-April 2023. The research was a two-factor factorial experiment designed in a Completely Randomized Design. The first factor was the explants of garlic shoots consisting of the lower, middle and upper shoots. The second factor was the size of the explants of garlic shoots consisting of 3 mm, 6 mm and 9 mm. The results showed that there was no interaction between the size of the explants and the size of the explants on the growth of the garlic plantlets. The lower shoots with sizes of 3mm, 6mm and 9mm were able to induce complete plantlets, the middle buds with sizes of 3mm, 6mm and 9mm produced complete plantlets, while the upper explants with sizes of 3mm, 6mm and 9mm not yet able to show complete plantlet growth.
