Abstract

The high dependence on synthetic chemical pesticides for pest control can cause various adverse effects, including pest resistance, pesticide residues, and disruption to the environment and non-target organisms. One environmentally friendly alternative for pest management is the utilization of entomopathogenic nematodes (EPNs). This study aimed to determine and compare mortality, infection symptoms, and the time required to cause 50% mortality of the population or *Lethal Time* 50 (LT₅₀) in larvae of *Spodoptera litura*, *Plutella xylostella*, and *Crocidolomia pavonana* following the application of *Steinernema* spp. Kediri isolate at a concentration of 800 IJ/mL. The study employed a Completely Randomized Design (CRD) involving three insect species, namely *S. litura*, *P. xylostella*, and *C. pavonana*, each subjected to control and *Steinernema* spp. suspension treatments at a concentration of 800 IJ/mL. Each treatment consisted of four replications with 10 larvae per replication, resulting in a total of 240 larvae. The nematodes were propagated *in vivo* using *Tenebrio molitor* larvae and harvested using the *white trap* method. A 5 mL nematode suspension was applied to each treatment container containing 10 third-instar larvae. Mortality was observed up to 120 hours after application and analyzed using ANOVA followed by Duncan’s Multiple Range Test (DMRT) at the 5% significance level, while LT₅₀ values were determined using probit analysis. The results showed that *Steinernema* spp. Kediri isolate caused mortality in all tested insect species. The highest mortality at 120 hours after application was observed in *P. xylostella* at 92.5%, followed by *C. pavonana* at 85.0% and *S. litura* at 80.0%. Infected larvae exhibited symptoms including cessation of feeding, weakness and softening of the body, and changes in body coloration. The shortest LT₅₀ was recorded in *P. xylostella* at 17.98 hours, whereas *S. litura* and *C. pavonana* had LT₅₀ values of 73.72 hours and 74.64 hours, respectively. These results indicate variation in the responses of the three larval species to *Steinernema* spp. infection, with *P. xylostella* showing the highest mortality and the shortest time required to cause 50% population mortality.
