Abstract
Anthracnose caused by Colletotrichum sp. is one of the major diseases reducing chili production. Seed coating using Bacillus sp. has been recognized as an environmentally friendly biological control strategy. Although Carboxymethyl Cellulose, Arabic gum, and sodium alginate have been used as adhesive materials in seed coating formulations, comparative information regarding their effectiveness in maintaining the viability and antagonistic activity of Bacillus sp. remains limited. Therefore, this study aimed to evaluate the effect of different adhesive materials on the viability and antagonistic activity of Bacillus sp. against Colletotrichum sp. This study employed a Completely Randomized Design (CRD) with four treatments: control (KO), Carboxymethyl Cellulose (SC), Arabic gum (SA), and sodium alginate (SN), each with six replications. Bacterial viability was determined using the Total Plate Count (TPC) method, while antagonistic activity was evaluated using the dual culture method. Data were analyzed using ANOVA followed by Duncan's Multiple Range Test (DMRT) at a 5% significance level. Adhesive materials significantly affected the viability and antagonistic activity of Bacillus sp. SC produced the highest bacterial viability, followed by SN, whereas SA showed the lowest viability. In the antagonistic assay, SN produced the largest inhibition zone (9.5 mm), followed by SA (5.1 mm) and SC (3.6 mm), while the control produced no inhibition zone. Sodium alginate was the most effective adhesive in enhancing the antagonistic activity of Bacillus sp. against Colletotrichum sp., whereas Carboxymethyl Cellulose better maintained bacterial viability.
