Abstract
Moringa oleifera leaves are a source of flavonoid compounds that have high antioxidant activity so they have the potential to be used as raw materials in the food, pharmaceutical, and health industries. This study aims to evaluate the effect of extraction time and the mole ratio of choline chloride and citric acid-based Natural Deep Eutectic Solvent (NADES) on extract yield, Total Flavonoid Content (TFC), and mass transfer characteristics in the flavonoid extraction process using the Ultrasound-Assisted Extraction (UAE) method. The extraction time variations used were 10, 20, 30, 40, and 50 minutes, while the mole ratio of choline chloride to citric acid included 1:0.5, 1:0.75, 1:1, 1:1.25, and 1:1.5. The extract obtained was analyzed for its yield value, then the total flavonoid content was determined using the AlCl₃ method with a UV-Vis spectrophotometer at a wavelength of 437 nm. In addition, the volumetric mass transfer coefficient (kLa) was calculated to describe the extraction kinetics. The results showed that variations in extraction time and NADES solvent composition affected the yield, total flavonoid content, and mass transfer characteristics. The highest yield of 30.1333% was obtained at a mole ratio of 1:1 with an extraction time of 50 minutes. The maximum TFC value of 76.3094 mg QE/g extract was obtained at the same mole ratio with an extraction time of 40 minutes. The highest equilibrium concentration (Cs) value was also produced by a mole ratio of 1:1, which indicates the solvent's ability to optimally dissolve flavonoids. Based on these results, a mole ratio of choline chloride and citric acid of 1:1 is the best condition for the extraction of moringa leaf flavonoids using the UAE method.
