Abstract
Oryza sativa L. is a major food commodity and a source of carbohydrates for most of Indonesia's population. Based on BPS data, there is a decline in rice production every year. This decline in rice production is caused by plant pest attacks (PPH). Xanthomonas oryzae is a bacterium that causes bacterial leaf blight (BLB) in rice, which results in crop losses of up to 60-80%. An environmentally friendly alternative in controlling Xanthomonas oryzae is by using endophytic bacteria Bacillus sp. and Pseduomonas fluorescens. The experiment was conducted with two tests: in vitro and in vivo tests. The in vitro test was arranged in a completely randomized design with 4 treatments and 4 replications, resulting in 16 experimental units. The experimental unit consisted of one petri dish containing Nutrient Agar (NA) media. Antagonistic activity was evaluated using the disc diffusion method against Xanthomonas oryzae. The in vivo test was arranged in a completely randomized factorial design with 10 treatments and 3 replications. Each replication consisted of six rice plants planted in polybags, resulting in a total of 180 plants as experimental units. Treatments consisted of varying concentrations of antagonistic bacterial suspensions applied to the soil prior to pathogen inoculation. The pathogen Xanthomonas oryzae was inoculated onto rice plants during the vegetative phase. Bacillus sp. and Pseudomonas fluorescens were shown to suppress Xanthomonas oryzae disease. Results showed that a consortium treatment of these two bacteria produced the highest inhibition rate of 9.125 mm. In vivo testing, the consortium treatment with 20 ml/plant resulted in the lowest disease intensity of 6.06% and an incubation period of 4 days. The consortium treatment of Bacillus sp. and Pseudomonas fluorescens was able to suppress bacterial leaf blight compared to other treatments.
