Abstract

The intensive use of propineb-based fungicides in agriculture can negatively affect the environment due to toxic and persistent residues in soil. An environmentally friendly approach to reduce these impacts is bioremediation using microorganisms such as Streptomyces sp. This study aimed to examine the tolerance and biodegradation potential of Streptomyces sp. against 70% propineb fungicide under in vitro conditions. The research was conducted at the Kesehatan Tanaman 1 Laboratory, Universitas Pembangunan Nasional “Veteran” East Java, from June to August 2025. The experiments were carried out in two stages: (1) tolerance tests on Glucose Nutrient Agar medium supplemented with propineb at five concentrations (0; 0,50; 1,00; 1,50; 2,00 g/L), and (2) degradation tests using a dual-culture method with Colletotrichum sp. as a bioindicator. Observation variables included colony counts of Streptomyces sp. determined by the Total Plate Count (TPC) method for tolerance testing, changes in absorbance values (Abs₀–Abs₇) as an indicator of medium turbidity, colony diameter of Colletotrichum sp., and relative inhibition level (THR) for degradation assessment. Results showed that Streptomyces sp. was able to grow at all treatments, with the highest population of 1,42×10⁴ CFU/mL, decreasing with higher concentrations to 3,22×10¹⁰ CFU/mL. Absorbance values increased in all treatments, with the highest ΔAbs (0,977) at 1.50 g/L. Colletotrichum sp. colonies ranged from 3,49 cm in the negative control to 2,24 cm at 2,00 g/L, while THR values were between 3,17% and 35,48%. These findings indicate that Streptomyces sp. can survive in propineb-containing media and fungicide concentration affects relative inhibition against Colletotrichum sp.
