Abstract

Watermelon (Citrullus lanatus) is one of the most popular fruits in Indonesia, one of which is seedless watermelon. The seedless watermelon is a triploid watermelon resulting from a cross between a diploid watermelon as a male and a tetraploid watermelon as a female. Efforts to increase the production of different types of watermelon require the use of genetic engineering technology using colchicine. The aim of the study was to determine the effect of colchicine concentration and soaking time on the morphological and agronomic characters of watermelon (Citrullus lanatus). The research was conducted in Sungegeneng Village Land, Sekaran District, Lamongan Regency. The research design used a factorial experiment consisting of 2 factors, namely the concentration and soaking time of colchicine which was designed using a randomized block design (RBD) with 4 repetitions. The first factor was the concentration of colchicine consisting of 5 levels namely 0.00%, 0.05%, 0.10%, 0.15%, 0.20%, soaking time as a factor consisting of 2 levels namely 12 hours, 24 hours, so that there were 10 treatment combinations. The results showed that the combination of 0.20% colchicine concentration and 24-hour soaking time gave the best results in the seedling phase, namely the length of the seedlings and the number of leaves. While in the plant phase there are parameters of plant length, number of leaves, fruit circle, seed size, seed diameter and weight of 100 seeds. The colchicine concentration of 0.20% gave the highest yields on stem diameter, fruit weight per fruit, fruit weight per plant and fruit weight per plot. Soaking time of 24 hours was able to give the highest results on the number of leaves at the age of flower emergence, fruit weight per fruit, fruit weight per plant, fruit weight per plot.
