Abstract

Beta carotene is the largest natural pigment found in the skin mango fruit, which is known as a precursor of vitamin A which is able to fight cancer cells, heart disease, reduce eye disease, antioxidants, and regulator in the immune system. Beta carotene can be extracted using multilevel maceration extraction method with two types of solvents namely n hexane and cyclohexane. This study aims to extract beta carotene from the skin arumanis mango and looking for the best extraction time and ratio of ingredients: solvent in producing beta carotene extract. Arumanis mango skin waste dried and mashed, obtained mango skin powder as much as 10 g then reacted with n-hexane solvent with the ratio of materials and solvents (1:6, 1:7, 1:8, 1:9, and 1:10 b/v), then macerated in an incubator at 35℃ with the time (20, 22, 24, 26, and 28 hours). Filtrate I was extracted again with the second solvent n-hexane and cyclohexane (50%:50% v/v) with the ratio of materials and same solvent and extraction time. The resulting filtrate I and II are mixed produce filtrate III which is then stored in the refrigerator. Solvent and Beta-carotene is separated by centrifugation. Extract carotene results from the skin. The mangoes obtained were then analyzed by UVVis spectrophotometry to get absorbance and wavelength data. Analysis results showed the best β-carotene content at the ratio of ingredients: solvent 1:10 and time 26 hours of extraction of 2.5137 mg/100g with an absorption (abs) of 3.3012 at wavelength 336 nm. The longer the maceration extraction time, the done and the greater the ratio of material: solvent can not always increase the level of β-carotene produced.
